Sample Collection
Discontinue heparin for 1 day and oral anticoagulants for 7 days with the treating physician's consent. Overnight fasting is preferred. Duly fill Chromosome and FISH Analysis Requisition Form (Form 17) and Coagulation Requisition Form (Form 15).
Method: Venipuncture
Laboratory Analysis
Collect samples from both husband and wife as specified. Mix Blue Top tube thoroughly by inversion. Avoid haemolysis and do not freeze Green Top tubes.
Report Delivery
Transport Green Top tubes refrigerated immediately, reaching lab within 24 hours. Blue Top tube to lab within 4 hours; if delayed, prepare platelet-poor plasma by double centrifugation at 3600 rpm for 15 minutes each and freeze immediately. SST serum ship refrigerated or frozen. Do not thaw frozen samples.

How to Prepare
- Collect 5 mL whole blood in 2 Green Top (Sodium Heparin) tubes each from husband and wife.
- Ship Green Top tubes refrigerated immediately and do not freeze; specimen must reach lab within 24 hours.
- Collect 3 mL whole blood in 1 Blue Top (Sodium Citrate) tube and mix thoroughly by inversion.
- Transport Blue Top tube to lab within 4 hours; if this is not possible, prepare platelet-poor plasma within 1 hour by centrifugation at 3600 rpm for 15 minutes, repeat centrifugation, transfer supernatant to a clean plastic vial, and freeze immediately.
- Collect 4 mL serum from 2 SSTs and ship refrigerated or frozen.
- Ensure Chromosome and FISH Analysis Requisition Form (Form 17) and Coagulation Requisition Form (Form 15) are duly filled.

Reduce the likelihood of antiphospholipid syndrome but do not exclude all causes

Limitations
- ⚠Karyotype cannot detect single-gene mutations or microdeletions below the resolution of standard cytogenetic analysis
- ⚠Positive antiphospholipid antibody results require clinical correlation and may need repeat testing as per guidelines
- ⚠Negative results do not rule out all causes of infertility or recurrent pregnancy loss
