Method: Venipuncture
Laboratory Analysis
A healthcare professional will collect approximately 2 mL of blood from a vein in your arm using standard venipuncture technique. The blood is drawn into a serum separator tube (SST). The procedure typically takes less than 5 minutes and may cause minimal discomfort.
Report Delivery
Apply pressure to the puncture site with a cotton ball or bandage for a few minutes. Mild bruising may occur. You can resume normal activities immediately. The sample is processed using agarose gel electrophoresis to separate and quantify individual CK isoenzymes.

How to Prepare
- Provide 2 mL (minimum 1.5 mL) of serum from one SST tube
- Hemolyzed specimens are NOT acceptable and will be rejected
- Ship the sample refrigerated (2–8°C) or frozen (≤ -20°C)
- Refrigerated samples are stable for 72 hours; frozen samples are stable for 1 week
- Avoid hemolysis by proper venipuncture technique and gentle mixing of the sample
- Clearly label the sample with patient details and date/time of collection

While troponin assays are now the gold standard for acute myocardial infarction, CK-MB fractionation via electrophoresis helps identify skeletal muscle disease, rhabdomyolysis, and mixed muscle injury patterns that can confound cardiac-only biomarkers. I recommend this test when there is discordance between clinical presentation and troponin levels, or when a non-cardiac source of CK elevation is suspected."
