Overview
The primary purpose of the Histopathology Skin Biopsy Panel Test is to provide a definitive microscopic diagnosis of skin conditions that cannot be confirmed through clinical examination alone. This test enables pathologists to examine skin tissue at the cellular level to detect cancerous or precancerous cells, identify inflammatory and autoimmune conditions, diagnose skin infections, assess margins of excised lesions, and grade and stage skin cancers. The results directly guide dermatologists and oncologists in formulating effective treatment plans, determining prognosis, and monitoring disease progression or treatment response.

Sample Collection
Brief clinical history in Histopathology Requisition Form (Form 2) is mandatory. Inform the dermatologist about any blood-thinning medications, allergies, or underlying medical conditions. No fasting is required for this test.
Method: Skin biopsy (incisional, excisional, punch, or shave biopsy as determined by the referring dermatologist)
Laboratory Analysis
A skin biopsy will be performed by the dermatologist. The area will be cleaned and local anaesthesia will be administered. A small piece of skin tissue will be removed using an appropriate biopsy technique (punch, incisional, excisional, or shave). The specimen will be placed immediately in 10% formal-saline. A bandage or suture will be applied to the biopsy site.
Report Delivery
Keep the biopsy site clean and dry. Follow the dermatologist's wound care instructions. Mild discomfort, slight bleeding, or swelling at the biopsy site is normal. Report signs of infection such as increasing redness, pus, or fever to your doctor. Results will be available in approximately 5 working days.
Timeline: Sample collection daily by 6 PM. Reports are available within 5 working days from sample receipt. Reports can be accessed via online portal, email, or WhatsApp.

How to Prepare
- Submit specimen in 10% Formal-saline
- Formal-saline should cover the specimen completely
- Ship at room temperature
- Brief clinical history in Histopathology Requisition Form (Form 2) is mandatory
- For patients with multiple specimens, register all specimens under a single lab number
- Label the specimen container with patient name, ID, site of biopsy, and date of collection
Doctor's Notes
Reviewed by Dr Pasupathy Arumugam — MBBS, MD (Pathology) · Reg. No. 21521
"A histopathology skin biopsy panel is one of the most reliable diagnostic tools available to dermatologists. It allows us to examine skin tissue at a cellular level, enabling accurate identification of skin cancers, autoimmune conditions, inflammatory dermatoses, and infections that may not be diagnosable through clinical examination alone. I routinely recommend this test for patients with persistent, atypical, or non-responsive skin lesions. Early and precise diagnosis through histopathology significantly improves treatment outcomes and patient prognosis."

Test Parameters & Specifications
Sample Stability
- Specimen received without formal-saline or in dried condition
- Specimen container not labelled or mislabelled
- Requisition form (Form 2) missing or incomplete clinical history
- Specimen too small or severely crushed for meaningful histological evaluation
- Specimen received in expired or inadequate formal-saline solution
Understanding Your Results
Normal skin architecture with no evidence of malignancy, significant inflammation, or infection. May include benign lesions such as dermatofibroma, seborrheic keratosis, or benign naevus.
Action: Discuss results with your dermatologist. No further intervention may be needed unless symptoms persist.
Presence of cancerous cells consistent with basal cell carcinoma, squamous cell carcinoma, melanoma, or other skin malignancies. Report may include tumour type, grade, depth of invasion, and margin status.
Action: Urgent consultation with dermatologist or oncologist required for staging, treatment planning, and possible additional procedures.
Inflammatory infiltrate pattern consistent with conditions such as psoriasis, eczema, lichen planus, lupus, or other autoimmune dermatoses.
Action: Consult your dermatologist for appropriate treatment and management plan.
Special stains or morphological features indicating fungal, bacterial (including AFB), or viral infection of the skin.
Action: Consult your dermatologist or infectious disease specialist for targeted antimicrobial therapy.
Atypical cells or dysplastic changes that may indicate actinic keratosis, Bowen's disease, or dysplastic naevus with potential for progression.
Action: Regular follow-up with dermatologist recommended. Additional excision or treatment may be advised.
Specimen insufficient for definitive diagnosis due to small size, crush artifact, or non-representative sampling.
Action: Repeat biopsy may be recommended by your dermatologist with adequate sampling of the lesion.
Consult your dermatologist or physician if you notice any new, changing, or persistent skin lesions; non-healing wounds; rapidly growing lumps; changes in the size, shape, or color of existing moles; skin lesions that bleed, itch, or crust over; or if your histopathology report indicates malignant, dysplastic, or inconclusive findings. Early consultation ensures timely intervention and better outcomes.
Limitations
- ⚠Histopathology provides a snapshot of the tissue at the time of biopsy; it may not represent the full extent of the disease
- ⚠Some conditions may require additional immunohistochemistry or molecular studies for definitive diagnosis
- ⚠Early-stage or superficial biopsies may not capture deeper pathology
- ⚠Overlap in histological patterns between different skin conditions may occasionally make definitive diagnosis challenging
Risks & Considerations
- ●Minor bleeding or bruising at the biopsy site
- ●Infection at the biopsy site (uncommon with proper wound care)
- ●Scarring at the biopsy site
- ●Mild pain or discomfort during and after the procedure
- ●Allergic reaction to local anaesthesia (rare)
- ●Need for repeat biopsy if the initial specimen is non-diagnostic
Interfering Factors
- ●Inadequate specimen preservation or improper fixation in formal-saline
- ●Specimen crush artifact or thermal damage during biopsy collection
- ●Insufficient clinical history provided on the requisition form
- ●Specimen too small or superficial for adequate tissue evaluation
- ●Delayed transportation of specimen to the laboratory

| Comparison | Histopathology Skin Biopsy Panel Test | Dermoscopy is a non-invasive surface examination of skin lesions using a dermatoscope. While it aids in initial evaluation, it cannot provide cellular-level detail like histopathology. Histopathology remains the gold standard for definitive diagnosis. | Skin cultures identify specific infectious organisms by growing them in the laboratory. Histopathology can detect tissue response to infection, identify organisms on staining, and diagnose non-infectious conditions that culture cannot. | Immunofluorescence is used specifically for autoimmune blistering diseases. It detects antibody deposits in skin tissue. Histopathology provides a broader evaluation and may be complemented by immunofluorescence when autoimmune conditions are suspected. |
