Loop Mediated Isothermal Amplification (LAMP), Qualitative DNA PCR
Step 1

Sample Collection

No special preparation is required. Please inform your doctor if you are taking anti-tubercular medications.

Method: Expectorated sputum collection / aseptic collection for invasive samples

Step 2

Laboratory Analysis

For sputum, you will be asked to cough deeply to bring up phlegm from the lungs. For other sample types, a physician or trained staff will collect the sample under sterile conditions.

Step 3

Report Delivery

The sample should be transported to the laboratory as soon as possible. If delay is expected, refrigerate at 2-8°C. Do not freeze.

Timeline: Samples submitted by 11 am are reported the same day. Reports are available online, by email or on WhatsApp.

Patient Instructions

1
Before the Test:No special preparation is required. The test can be done at any time of the day; however, first morning sputum sample is preferred.
2
During the Test:You may be asked to cough deeply to produce sputum. If another sample type is required, a trained professional will collect it under sterile conditions.
3
After the Test:No specific precautions are needed after sample collection. Your report will be delivered online on the same day if the sample reaches the lab by 11 am.

About This Test

Who Should Get This Test

To detect the presence of Mycobacterium tuberculosis complex DNA in respiratory and selected extrapulmonary samples to aid in the diagnosis of tuberculosis.

How to Prepare

  • Collect first morning sputum after deep cough into a sterile screw-capped container.
  • Ensure the sample is from the lower respiratory tract, not saliva.
  • Ship refrigerated at 2-8°C. DO NOT FREEZE.
  • For body fluids, tissue, urine or bronchial samples, follow the collection instructions given by the laboratory or physician.

Understanding Your Results

The TB LAMP test provides a qualitative result for M. tuberculosis complex DNA. The result should be interpreted in the clinical context and with other laboratory and radiological findings.
Positive: M. tuberculosis complex DNA detected. This supports the diagnosis of tuberculosis and requires clinical correlation and appropriate anti-TB treatment if not already started.
Negative: M. tuberculosis complex DNA not detected. This does not completely exclude tuberculosis; results should be interpreted with clinical, radiological and culture findings.
Combining TB LAMP with TB culture increases overall diagnostic sensitivity, especially when clinical suspicion remains high.
⚠️ When to Consult a Doctor:

Consult a physician if you have cough lasting more than 3 weeks, chest pain, blood-stained sputum, unexplained fever, night sweats or unintentional weight loss. Early diagnosis and treatment prevent spread and improve recovery outcomes.

Limitations

  • Qualitative test only; does not provide bacterial load.
  • Does not detect drug resistance.
  • A negative result does not exclude tuberculosis; culture remains important.
  • Whole blood sample is not acceptable for this test.

Risks & Considerations

  • No significant risk for sputum collection.
  • If invasive sample collection such as bronchoscopy is needed, there is a small risk of bleeding or infection at the sample site.

Interfering Factors

  • Improper sample collection with excess saliva may reduce detection.
  • PCR or LAMP inhibitors in clinical samples may rarely affect amplification.
  • Contamination during collection or processing can cause false-positive results.
  • Detection may be affected in patients already on anti-tubercular therapy.

Frequently Asked Questions

What is the TB LAMP Qualitative DNA PCR Mycobacterium Tuberculosis Complex Test?
It is a molecular diagnostic test that detects DNA of Mycobacterium tuberculosis complex in clinical samples using loop mediated isothermal amplification (LAMP). It is WHO-endorsed and helps in rapid diagnosis of tuberculosis.
What is the cost of the TB LAMP test at DNA Labs India?
The test costs Rs 5000, and DNA Labs India offers free home sample collection for online bookings.
What sample is needed for the TB LAMP test?
The preferred sample is 1-2 mL of first morning sputum. Other accepted samples include induced sputum, bronchial lavage, bronchial brushing, endotracheal secretions, gastric lavage, body fluids, tissue biopsy, and urine in specified volumes.
Is fasting required before the test?
No, no special preparation or fasting is required for the TB LAMP test.
How soon will I get my report?
If the sample is submitted by 11 am, the report is delivered on the same day via online portal, email or WhatsApp.
What does a positive TB LAMP result mean?
A positive result indicates detection of Mycobacterium tuberculosis complex DNA in the sample, supporting a diagnosis of tuberculosis. Your physician will correlate this with clinical symptoms and other tests.
What does a negative TB LAMP result mean?
A negative result means no M. tuberculosis complex DNA was detected in the tested sample. It does not completely rule out tuberculosis; clinical correlation and culture may be needed.
Is the TB LAMP test recommended by WHO?
Yes, WHO published a policy statement in 2016 endorsing TB LAMP as a rapid molecular test for diagnosis of pulmonary tuberculosis. It is also included in ICMR's draft essential test list.
Can the TB LAMP test detect drug-resistant tuberculosis?
No, TB LAMP is designed for qualitative detection of M. tuberculosis complex DNA and does not provide drug sensitivity information. Tests like CBNAAT are used for rifampicin resistance detection.
Can the test be done on blood samples?
No, whole blood is not acceptable. Valid samples include sputum, body fluids, tissue biopsy, urine, bronchial lavage, and other specified respiratory or sterile samples.
Do I need a doctor's prescription for this test?
It is recommended to consult a physician before the test, as results need clinical correlation. You can book the test for home sample collection and our physicians can guide you.
How should the sample be transported?
The sample should be refrigerated at 2-8°C and transported quickly. It must not be frozen. Room temperature stability is only 2 hours.
