Method: Expectorated sputum collection / aseptic collection for invasive samples
Laboratory Analysis
For sputum, you will be asked to cough deeply to bring up phlegm from the lungs. For other sample types, a physician or trained staff will collect the sample under sterile conditions.

The sample should be transported to the laboratory as soon as possible. If delay is expected, refrigerate at 2-8°C. Do not freeze.

To detect the presence of Mycobacterium tuberculosis complex DNA in respiratory and selected extrapulmonary samples to aid in the diagnosis of tuberculosis.
How to Prepare
- Collect first morning sputum after deep cough into a sterile screw-capped container.
- Ensure the sample is from the lower respiratory tract, not saliva.
- Ship refrigerated at 2-8°C. DO NOT FREEZE.
- For body fluids, tissue, urine or bronchial samples, follow the collection instructions given by the laboratory or physician.

Sample Stability
- Whole blood sample is not acceptable.
- Sample received frozen.
- Inadequate sample volume.
- Sample in a non-sterile or leaking container.
- Improperly labelled or unlabeled sample.

Limitations
- ⚠Qualitative test only; does not provide bacterial load.
- ⚠Does not detect drug resistance.
- ⚠A negative result does not exclude tuberculosis; culture remains important.
- ⚠Whole blood sample is not acceptable for this test.
Risks & Considerations
- ●No significant risk for sputum collection.
- ●If invasive sample collection such as bronchoscopy is needed, there is a small risk of bleeding or infection at the sample site.
Interfering Factors
- ●Improper sample collection with excess saliva may reduce detection.
- ●PCR or LAMP inhibitors in clinical samples may rarely affect amplification.
- ●Contamination during collection or processing can cause false-positive results.
- ●Detection may be affected in patients already on anti-tubercular therapy.
