Sample Collection
Ensure the duly filled Test Send Out Consent Form (Form 35) is completed prior to sample collection. Inform the phlebotomist about any current medications, recent immunoglobulin therapy, or blood transfusions. No fasting is required for this test.
Method: Venipuncture
Laboratory Analysis
A trained phlebotomist will collect approximately 2 mL of blood via venipuncture from a vein in the arm. The blood will be collected into a Serum Separator Tube (SST). The process typically takes less than 5 minutes and may involve minimal discomfort at the puncture site.
Report Delivery
After sample collection, gentle pressure will be applied to the puncture site with a cotton ball or bandage. Mild bruising may occur and typically resolves within a few days. The serum sample will be separated, processed, and shipped under refrigerated or frozen conditions to the testing facility.

How to Prepare
- Complete the Test Send Out Consent Form (Form 35) — this is mandatory for sample processing
- No fasting is required prior to sample collection

"IgA subclasses testing is clinically important in the evaluation of patients presenting with recurrent infections, suspected immunodeficiency, and IgA nephropathy. IgA nephropathy is the most common primary glomerulonephritis worldwide and is typically associated with aberrantly glycosylated IgA1. Quantifying IgA1 and IgA2 separately allows nephrologists and immunologists to distinguish between mucosal and systemic IgA involvement, assess subclass-specific deficiencies, and evaluate risk for adverse transfusion reactions. This test, performed via nephelometry, provides precise quantitative results and should always be interpreted alongside clinical findings, family history, and complementary laboratory investigations such as total immunoglobulin levels and renal function panels."

Understanding Your Results
All IgA subclass levels are within the expected reference ranges, suggesting adequate IgA-mediated immune function. Clinical correlation is recommended if symptoms persist.
May indicate selective IgA1 subclass deficiency, which is associated with recurrent sinopulmonary infections and gastrointestinal infections. Further immunological workup is recommended.
May indicate IgA2 subclass deficiency, which can affect mucosal immunity particularly in the gastrointestinal tract. Increased susceptibility to mucosal infections may be observed.
Complete IgA subclass deficiency may indicate a more significant immunodeficiency. Patients may be at increased risk for recurrent infections, autoimmune disorders, and anaphylactic transfusion reactions.
Elevated IgA1, particularly aberrantly glycosylated IgA1, may be associated with IgA nephropathy — the most common primary glomerulonephritis. Nephrology evaluation and renal biopsy may be warranted.
Elevated total IgA may be seen in chronic liver disease, chronic infections, autoimmune conditions, or IgA myeloma. Clinical correlation with liver function tests and serum protein electrophoresis is advised.
Consult a nephrologist or immunologist if your IgA subclass levels are outside the reference range, if you experience recurrent respiratory or gastrointestinal infections, if you have a family history of immunodeficiency, or if you have been diagnosed with or are suspected to have IgA nephropathy. Early evaluation and management can help prevent complications such as chronic kidney disease, autoimmune disorders, and severe allergic reactions.
