Sample Collection
The tissue biopsy or surgical specimen must be properly fixed in 10% formal-saline or processed into a formalin-fixed paraffin-embedded (FFPE) block prior to submission. A copy of the histopathology report must be provided along with the site of biopsy and relevant clinical history. No special patient preparation such as fasting is required for this test as it is performed on an already collected tissue specimen.
Method: Tissue Biopsy / Surgical Excision
Laboratory Analysis
The tissue sample is collected via biopsy (excisional, incisional, or core needle) or surgical excision by the treating physician. The specimen is immediately placed in 10% neutral buffered formalin for fixation. The tissue is then processed, embedded in paraffin, and sectioned onto glass slides for immunohistochemical staining in the laboratory.
Report Delivery
After the immunohistochemical staining is completed, the stained slides are examined by a qualified pathologist who interprets the CD5 expression pattern, intensity, and percentage of positive cells. The results are compiled into a detailed report and made available within 5 to 7 working days via online portal, email, or WhatsApp.

How to Prepare
- Submit tumor tissue in 10% Formal-saline or as a Formalin Fixed Paraffin Embedded (FFPE) block
- Ship the specimen at room temperature
- Provide a copy of the histopathology report with the specimen
- Indicate the site of biopsy on the requisition form
- Provide relevant clinical history including suspected diagnosis
- Ensure tissue is fixed for a minimum of 6 hours and not more than 72 hours in formalin

Understanding Your Results
Aberrant CD5 expression on B cells is characteristic of mantle cell lymphoma (MCL) and chronic lymphocytic leukemia/small lymphocytic lymphoma (CLL/SLL). Further testing with Cyclin D1, CD23, and SOX11 is recommended to differentiate between these entities.
CD5 expression on T cells is a normal finding. However, loss of CD5 expression on T cells may suggest a T-cell lymphoma such as peripheral T-cell lymphoma not otherwise specified (PTCL-NOS) or anaplastic large cell lymphoma.
Absence of CD5 on B cells suggests a B-cell lymphoma other than MCL or CLL/SLL, such as follicular lymphoma, marginal zone lymphoma, or diffuse large B-cell lymphoma (most cases). Further panel testing is required for definitive classification.
Loss of CD5 on T cells is abnormal and may indicate certain T-cell lymphomas. Correlation with additional T-cell markers (CD2, CD3, CD7) and clinical context is essential.
You should consult an oncologist or hematologist if your biopsy results show abnormal CD5 expression, if you have been diagnosed with or are being evaluated for a lymphoproliferative disorder, or if you experience symptoms such as persistent unexplained lymphadenopathy, unexplained weight loss, night sweats, fever, or fatigue. Early consultation ensures timely diagnosis and initiation of appropriate treatment.
Limitations
- ⚠CD5 alone is not diagnostic and must be interpreted with a panel of immunohistochemical markers
- ⚠Cannot distinguish between reactive and neoplastic T-cell populations without additional markers
- ⚠Rare cases of CD5-negative mantle cell lymphoma or CD5-positive diffuse large B-cell lymphoma exist
- ⚠Does not provide information about specific genetic translocations such as t(11;14) in MCL
- ⚠Results should always be correlated with clinical findings, morphology, and flow cytometry data
