Sample Collection

Overnight fasting is preferred. A duly filled Coagulation Requisition Form (Form 15) is mandatory. It is recommended that the patient discontinues Heparin for 1 day and oral anticoagulants for 7 days before sampling, but only with the consent of the treating physician. Do not stop any prescribed medication without medical advice.

Laboratory Analysis

A phlebotomist will collect 3 mL of whole blood into a Blue Top sodium citrate tube and 5 mL of whole blood into two Lavender Top EDTA tubes. The citrate tube should be mixed gently by inversion to ensure proper anticoagulation.

Step 3

Report Delivery

The citrate tube must be transported to the laboratory within 4 hours. If this is not possible, platelet-poor plasma should be prepared within 1 hour of collection and frozen immediately. EDTA tubes should be shipped refrigerated, not frozen. Resume prescribed medications only on the treating physician's advice.

How to Prepare

  • Sample type: 3 mL whole blood in 1 Blue Top (Sodium Citrate) tube. Mix thoroughly by inversion.
  • Transport to lab within 4 hours.
  • If transport is delayed, make platelet-poor plasma (PPP) within 1 hour of collection: centrifuge at 3600 rpm for 15 minutes, transfer supernatant, centrifuge again, transfer to labelled plastic screw-capped vial and freeze immediately. Do not thaw.
  • EDTA sample: 5 mL (3 mL minimum) whole blood in 2 Lavender Top (EDTA) tubes. Ship refrigerated. Do not freeze.
  • Overnight fasting is preferred and Coagulation Requisition Form (Form 15) must be duly filled.
