Overview
The purpose of the Cell Line Authentication DNA Test is to verify the identity and purity of cell lines used in research and clinical applications. It detects misidentification, cross-contamination, and genetic alterations that may occur during cell culture. The test uses STR profiling to generate a unique DNA fingerprint of the cell line, which is then compared to reference databases (ATCC, DSMZ) for confirmation. This ensures that the cell line is authentic and suitable for experiments, clinical trials, or bioproduction.

Sample Collection
Follow the specific sample preparation instructions provided. For extracted DNA, ensure it is eluted in low TE buffer or nuclease-free water at a concentration of 10-100 ng/µl. For cell pellets, provide at least 1x10^6 cells/ml. For FTA spots, follow standard guidelines and spot 20 µl of sample. For tissue/tumor, provide approximately 5 mg.
Method: Home collection or lab visit
Laboratory Analysis
Samples should be collected in sterile, DNase-free containers. For FTA cards, use a single card per sample and pack in a separate zip bag. Ensure proper labeling with sample ID and date.
Report Delivery
Transport samples according to the specified conditions: extracted DNA and cell pellets should be frozen at -20°C and -80°C respectively, and shipped with ice packs or dry ice. FTA cards can be transported at room temperature. Tissue samples should be transported with ice packs.
Timeline: Reports are typically available within 10-12 days after sample receipt.

How to Prepare
- Extracted DNA: Elute in low TE buffer or nuclease-free water, 10-100 ng/µl, volume 10-20 µl. Freeze at -20°C and transport with ice pack.
- Cell Pellets: Provide 1x10^6 cells/ml. Freeze at -80°C and pack with dry ice.
- FTA Spot: Follow standard guidelines, spot 20 µl sample on the card. Pack single FTA card in a single zip bag.
- Tissue/Tumor: Size approx 5 mg. Transport with ice pack.

Test Parameters & Specifications
Sample Stability
- Insufficient quantity of DNA (<10 ng/µl) or cells (<1x10^6 cells/ml)
- Hemolyzed or degraded samples
- Improper labeling or documentation
- Samples not transported under required temperature conditions
Understanding Your Results
Match with reference profile
Cell line is authentic and suitable for research use.
No match with any reference profile
Cell line may be misidentified or novel; further investigation required.
Mixed profile (multiple alleles at multiple loci)
Indicates cross-contamination with another cell line; subcloning or re-authentication needed.
If you are a researcher or clinician and have concerns about the authenticity of your cell lines, or if you require guidance on interpreting the results, consult with a qualified geneticist or research director.
Limitations
- ⚠Test cannot detect all types of genetic mutations
- ⚠Reference database may not include all cell lines
- ⚠Results depend on sample quality and quantity
- ⚠Not intended for diagnostic or therapeutic use
Risks & Considerations
- ●No significant risks associated with sample collection
- ●Potential for inaccurate results if sample quality is poor
Interfering Factors
- ●Degraded DNA due to improper storage or transport
- ●Insufficient quantity of DNA or cells
- ●Contamination with other cell lines during sample collection
