Overview
This targeted NGS test identifies mutations in the RPL10 gene on the X chromosome. It is used to confirm a diagnosis of autism susceptibility, X-linked type 5, in affected males, and to detect carrier status in females. The test supports genetic counseling and helps assess recurrence risk in the family.
- Test Code
- 3910
- Price
- ₹20,000
- Sample Type
- Blood or Extracted DNA or One drop Blood on FTA Card
- Result Time
- Reports are delivered within 3 to 4 weeks after the sample reaches the laboratory.
- Fasting Required
- No
- Method
- Next Generation Sequencing (NGS)
Sample Collection
No fasting or special dietary preparation is required. We recommend completing a genetic counseling session before sample collection to document family history and draw a pedigree.
Method: Peripheral blood draw or finger-prick blood spot on FTA card
Laboratory Analysis
A small amount of blood will be collected from a vein in the arm, or a finger-prick sample can be placed on an FTA card. The procedure is quick and no anesthesia is required.
Report Delivery
There are no restrictions after sample collection. Results are expected within 3 to 4 weeks. DNA Labs India will share the clinical report and the raw data files.
Timeline: Reports are delivered within 3 to 4 weeks after the sample reaches the laboratory.

About This Test
Who Should Get This Test
This targeted NGS test identifies mutations in the RPL10 gene on the X chromosome. It is used to confirm a diagnosis of autism susceptibility, X-linked type 5, in affected males, and to detect carrier status in females. The test supports genetic counseling and helps assess recurrence risk in the family.
How to Prepare
- Please carry the signed test requisition form and a valid photo ID.
- For FTA cards, allow the blood spot to air-dry completely before sealing the pouch.
- Do not write directly on the FTA card blood spot.
- For whole blood sample, use an EDTA tube and gently mix the tube after collection.
Doctor's Notes
Reviewed by Dr Pasupathy Arumugam — MBBS, MD (Pathology) · Reg. No. 21521
"The RPL10 gene is on the X chromosome and mutations can lead to X-linked type 5 autism susceptibility. Testing should be done after detailed genetic counseling because results have implications for the patient and family. An NGS-based approach is useful to identify small sequence changes in RPL10 and guide recurrence-risk counseling."

Understanding Your Results
Pathogenic variant detected
The result is consistent with RPL10-related autism susceptibility, X-linked type 5. Clinical correlation and family studies are recommended.
Likely pathogenic variant detected
The variant is highly likely to be disease-causing. Additional family testing may be needed to confirm segregation.
Variant of uncertain significance (VUS)
The clinical significance of the variant is not known. Further testing of affected and unaffected family members may help clarify its role.
No pathogenic variant detected
This reduces the likelihood of RPL10-linked autism but does not exclude other genetic or non-genetic causes.
Consult a clinical geneticist, neurologist, or pediatrician if a pathogenic or likely pathogenic RPL10 variant is detected, or if autism symptoms are present and the cause has not been established.
Limitations
- ⚠This test analyzes only the RPL10 gene, not all autism-related genes.
- ⚠Large deletions, duplications, or complex structural variants may not be reliably detected by this targeted NGS test.
- ⚠Deep intronic variants or repeat expansions may not be covered.
- ⚠A negative result does not exclude other genetic or non-genetic causes of autism.
- ⚠Variants of uncertain significance may require additional family studies.
Risks & Considerations
- ●Minimal discomfort at the needle site
- ●Very small risk of bruising or infection
- ●No serious medical risks associated with genetic testing
Interfering Factors
- ●Poor DNA quality or quantity
- ●Sample contamination or sample mix-up
- ●Low-level mosaicism that may fall below the NGS detection threshold
- ●Large structural rearrangements not detected by standard NGS
- ●Incomplete family history limiting variant interpretation
