Doctor's Notes

Reviewed by — MBBS, MD (General Medicine) · Reg. No. 8052

"Flow cytometry immunophenotyping is essential for detecting the characteristic hairy cell leukemia phenotype, including CD11c, CD25, CD103 and CD123 co-expression. Results should always be correlated with morphology, blood counts and clinical findings."

Understanding Your Results

Flow cytometry results are reported as an immunophenotypic profile. Hairy cell leukemia cells typically show bright CD11c, CD25, CD103 and CD123 expression with mature B-cell markers such as CD19, CD20 and CD22. Results should be interpreted by the treating oncologist or hematologist in conjunction with morphology and clinical findings.
Positive hairy cell phenotype supports the diagnosis if clinical and morphological features are consistent
Loss or reduction of abnormal cells after treatment suggests response
Persistent abnormal population may indicate residual disease
Any immunophenotypic abnormality should be correlated with bone marrow and peripheral blood findings
⚠️ When to Consult a Doctor:

Consult your haematologist or oncologist if the report indicates an abnormal immunophenotype, or if you have symptoms such as fever, fatigue, weight loss, bleeding or recurrent infections.

Limitations

  • Flow cytometry immunophenotyping should be interpreted with morphology and clinical history
  • A negative result does not exclude disease if sampling is inadequate
  • Marker expression can vary after treatment
  • Bone marrow aspirate smear should be reviewed alongside flow cytometry

Risks & Considerations

  • Pain or bruising at venipuncture site
  • Bone marrow aspiration may cause local soreness

Interfering Factors

  • Refrigerated or frozen samples may reduce cell viability and marker expression
  • Clotted or haemolysed samples
  • Insufficient cell count or low tumour burden
  • Recent chemotherapy or immunotherapy may alter antigen expression
  • Delayed transport beyond 24 hours at room temperature
