Overview
The purpose of the Coagulation Profile 1 Test is to assess the overall clotting function of blood. Sample Collection
Overnight fasting is preferred. Ensure that a duly filled Coagulation Requisition Form (Form 15) is completed and accompanies the sample. Inform the phlebotomist about any medications, especially anticoagulants, that you are currently taking. Avoid alcohol consumption for 24 hours before the test.
Method: Venipuncture
Laboratory Analysis
Blood will be drawn via venipuncture into two separate tubes: one Lavender Top (EDTA) tube and one Blue Top (Sodium Citrate) tube. The Blue Top tube must be mixed thoroughly by gentle inversion immediately after collection. The phlebotomist will ensure correct labeling and proper fill volumes.
Report Delivery
Apply pressure to the puncture site for 3-5 minutes to prevent bruising. Avoid heavy lifting or strenuous activity with the affected arm for a few hours. The EDTA tube should be shipped refrigerated (do not freeze). The Sodium Citrate tube must be transported to the lab within 4 hours. If this is not possible, platelet-poor plasma (PPP) must be prepared within 1 hour of collection.

How to Prepare
- Collect 3 mL (1.5 mL min.) whole blood in 1 Lavender Top (EDTA) tube. Ship refrigerated. DO NOT FREEZE.
- Collect 3 mL whole blood in 1 Blue Top (Sodium Citrate) tube. Mix thoroughly by inversion immediately.
- Transport the Sodium Citrate sample to the lab within 4 hours of collection.
- If transport within 4 hours is not possible, prepare PPP within 1 hour: centrifuge at 3600 rpm for 15 min, transfer supernatant to a clean plastic tube, centrifuge again at 3600 rpm for 15 min, then transfer supernatant (PPP) to a labelled screw-capped vial. FREEZE IMMEDIATELY. Ship frozen. DO NOT THAW.
- Duly filled Coagulation Requisition Form (Form 15) is mandatory.
- Overnight fasting is preferred prior to sample collection.

Understanding Your Results
May indicate vitamin K deficiency, liver disease, DIC, or the effect of warfarin therapy. Further investigation with specific factor assays may be needed.
May suggest deficiencies in intrinsic pathway clotting factors (e.g., Factor VIII, IX, XI, or XII), heparin therapy, or lupus anticoagulant. Common in hemophilia A and B.
May indicate fibrinogen deficiency, dysfibrinogenemia, or the presence of heparin in the sample. Elevated levels of fibrin degradation products can also prolong TT.
May suggest platelet disorders (e.g., thrombocytopenia, von Willebrand disease), vascular abnormalities, or the effect of antiplatelet medications such as aspirin.
May indicate severe deficiencies in clotting factors, significant thrombocytopenia, or the presence of anticoagulants in the blood.
Low platelet count (thrombocytopenia) may contribute to prolonged bleeding. Low hemoglobin may indicate chronic blood loss from an undiagnosed bleeding disorder.
Consult your doctor if your Coagulation Profile 1 Test results show any abnormal parameters, if you experience symptoms such as unexplained bruising, prolonged bleeding from cuts, heavy menstrual periods, frequent nosebleeds, blood in urine or stool, or joint pain and swelling.
