Overview
The FISH - Eosinophilic Leukemia Panel is designed to detect specific genetic mutations associated with eosinophilic leukemia. It targets genes including PDGFRA (4q12), PDGFRB (5q33), FGFR1 (8p12), and CBFB (inv16). Identifying these abnormalities helps in confirming the diagnosis, differentiating from other conditions, predicting disease progression, and selecting appropriate targeted therapies such as tyrosine kinase inhibitors.

- No
- Method
- Fluorescence In Situ Hybridization (FISH)
Sample Collection
Ensure the Chromosome & FISH analysis Requisition Form (Form 17) is duly filled. No fasting required. Inform the healthcare provider about any medications or recent treatments.
Method: Venipuncture for blood; bone marrow aspiration for bone marrow
Laboratory Analysis
For blood sample: Venipuncture will be performed to collect 5 mL of blood into Sodium Heparin tubes. For bone marrow: Aspiration will be done to collect 4 mL of marrow. The process is quick and may cause minimal discomfort.
Report Delivery
Apply pressure to the puncture site to prevent bleeding. Sample is shipped at 18-22°C; do not freeze. Avoid strenuous activity if bone marrow was aspirated. Results will be available in 4 working days.
Timeline: 4 Working days from sample receipt

How to Prepare
- Duly filled Chromosome & FISH analysis Requisition Form (Form 17) is mandatory
- Use 2 Green Top (Sodium Heparin) tubes for sample collection
- Ship at 18-22°C; do not freeze
- Label samples with patient details and test name
- For bone marrow collection, follow sterile technique

Test Parameters & Specifications
Sample Stability
- Sample not collected in Sodium Heparin tubes
- Sample frozen or stored at incorrect temperature
- Insufficient sample volume
- Missing or incomplete Requisition Form (Form 17)
- Hemolyzed or contaminated sample
Understanding Your Results
Positive for PDGFRA rearrangement
Indicates eosinophilic leukemia associated with PDGFRA mutation; may respond to specific TKIs like imatinib
Positive for PDGFRB rearrangement
Suggests eosinophilic leukemia linked to PDGFRB mutation; targeted therapy options available
Positive for FGFR1 rearrangement
Associated with aggressive eosinophilic leukemia; prognosis may vary
Positive for CBFB (inv16)
Indicates acute myeloid leukemia with eosinophilia; treatment may differ
Negative for all abnormalities
No detected mutations; further tests may be needed if clinical suspicion remains
Consult a doctor if you experience symptoms such as persistent fatigue, unexplained fever, weight loss, enlarged lymph nodes, or night sweats. Also, seek medical advice if your blood tests show high eosinophil counts or if you have a family history of eosinophilic disorders. After receiving test results, discuss them with an oncologist for appropriate management.
Limitations
- ⚠This test detects only specified genetic abnormalities; other mutations may not be identified
- ⚠False negatives can occur if the sample has insufficient tumor cells
- ⚠Not a standalone diagnostic tool; must be correlated with clinical and pathological findings
- ⚠Limited to eosinophilic leukemia-associated genes; does not cover all leukemia types
Risks & Considerations
- ●Mild pain or bruising at blood collection site
- ●Rare risk of infection or excessive bleeding from bone marrow aspiration
- ●Minimal discomfort during procedure
Interfering Factors
- ●Contaminated or improperly stored samples
- ●Use of fixatives other than Sodium Heparin
- ●Low cellularity in bone marrow samples
- ●Recent chemotherapy or radiation therapy affecting cell DNA
